Большой интерес представляет изучение комплексов Aβ с лекарственными средствами, которые рассматриваются в качестве перспективных кандидатов для терапии болезни Альцгеймера. К данной категории относится группа гипогликемических препаратов, в т. ч. инсулин детемир, аналог инсулина длительного действия. В данной работе методом биослойной интерферометрии впервые получены кинетические и равновесные параметры взаимодействия инсулина детемир с мономерными формами Aβ с длиной цепи 40 и 42 аминокислоты, а также предложена модель комплексов «инсулин детемир – Аβ40/42». Полученные данные могут быть использованы для изучения механизмов влияния инсулина детемир на когнитивные функции пациентов c диагнозом болезнь Альцгеймера.</p></abstract><trans-abstract xml:lang="en"><p>The study of β-amyloid peptide (Aβ) interactions with protein and peptide ligands is a relevant research task. Aβ complexes with drugs that are considered as promising candidates for the therapy of Alzheimer’s disease are of particular interest. This category includes a group of hypoglycemic drugs, such as insulin detemir, a long-acting insulin analogue. In this work, the method of biolayer interferometry is used to obtain the kinetic and equilibrium parameters of interaction of insulin detemir and monomeric Aβ forms with a chain length of 40 and 42 amino acids. A model of “insulin detemir–Aβ40/42” complexes is proposed.
