Это явление получило обозначение «химический мозг». Исследование закономерностей индукции «химического мозга» в эксперименте и его моделирование являются основой для последующей разработки фармакологических средств профилактики и коррекции. Цель работы — исследование дозовых эффектов при курсовом введении доксорубицина на долговременную пространственную память и микроскопическую картину префронтальной коры и гиппокампа у крыс. Доксорубицин вводили беспородным белым крысам-самцам (n=40) в дозах 2, 3 и 5 мг/кг, внутрибрюшинно, на 1, 7, 14 и 21-й день исследования. Животные контрольной группы получали изотонический раствор натрия хлорида. На 24-й и 25-й день выполняли оценку функции памяти в тесте «Y-лабиринт», на 32-й день — патолого-анатомическое вскрытие для последующего морфологического исследования структур головного мозга. У крыс, получавших доксорубицин, обнаружено зависимое от дозы снижение времени, проводимого в «новом» рукаве Y-лабиринта, что указывает на ослабление пространственной памяти. Микроскопическое исследование показало нарушение церебральной микроциркуляции в префронтальной коре, а также зонах СА1 и СА3 гиппокампа только после использования доксорубицина в максимальной дозе 5 мг/кг. Полученные результаты свидетельствуют, что при моделировании состояния «химический мозг» доксорубицином предпочтительно использовать препарат в дозе 2 мг/кг, вызывающей когнитивные нарушения при минимальном токсическом эффекте. Таким образом, экспериментально обоснован выбор дозы доксорубицина и теста для оценки нарушения пространственной памяти, предпочтительные при моделировании «химического мозга» в рамках проведения поиска и доклинического изучения средств фармакологической коррекции побочных эффектов химиотерапии.</p></abstract><trans-abstract xml:lang="en"><p>Despite the clear successes of chemotherapy in oncology, a significant rise is observed in cognitive impairment during pharmacotherapy and recovery — a phenomenon known as chemobrain. Experimental study of mechanisms underlying chemobrain induction and its modeling provide a critical foundation for developing pharmacological strategies for prevention and correction. The study aimed to evaluate the dosedependent effects of doxorubicin administration on long-term spatial memory and the microscopic morphology of the prefrontal cortex and hippocampus in rats. Doxorubicin was administered intraperitoneally to outbred male white rats (n=40) at doses of 2, 3, and 5 mg/kg on days 1, 7, 14, and 21. Control animals received isotonic saline. Memory function was assessed using the Y-maze test on days 24 and 25, followed by a morphological examination of brain structures on day 32. Rats treated with doxorubicin exhibited a dose-dependent decrease in the time spent in the novel arm of the Y-maze, indicating impaired spatial memory. Microscopic examination revealed disrupted cerebral microcirculation in the prefrontal cortex, as well as in the hippocampal CA1 and CA3 regions (at the maximum dose of 5 mg/kg). These findings suggest that for modeling chemobrain, a doxorubicin dose of 2 mg/kg is optimal, as it induces cognitive impairment with minimal toxicity.
