Линейные фрагменты плазмидных генно-инженерных конструкций, включающих нуклеотидные последовательности генов NAT1 и NAT2 человека под промотором гена альбумина мыши ( hNAT1 и hNAT2), микроинъецировали в мужские пронуклеусы зигот, полученных от самок F1 гибридных мышей (CBA/lac * C57BL/6). При подготовке самок-реципиентов также был использован приём предварительной подсадки самок-реципиентов к вазэктомированным самцам через перегородку, который способствовал стимуляции процесса фолликулогенеза и синхронизации половых циклов у самок-реципиентов. Это позволило получать большее количество псевдобеременных самок-реципиентов с копуляционными пробками к определённому времени, что является одним из критериев полноценности животного-реципиента и возможности успешной трансплантации ему эмбрионов, микроинъецированных генно-инженерными конструкциями. На основе этих приёмов разработан вариант модифицированной технологии получения трансгенных мышей с генами NAT1 и NAT2 человека.</p></abstract><trans-abstract xml:lang="en"><p>The method to induce superovulation in mice-donors, including the synchronization of sexual cycles of the female donor by pre-infusion to males through the partition, which allows you to get to determine time up to 17 zygotes per donor with highly visible pronucleus, suitable for introduction in them of genetically engineered structures was improved. It is possible to obtain a greater number of female pseudoharmonic-recipients with copulation wads to determine time, which is one of the criteria of usefulness of the animal recipient and the possibility of successful transplantation of the embryos to it, microinjection genetically engineered constructs.
